چکیده

objective: boron (b) is essential for plant development and might be an essential micronutrient for animals and humans. this study was conducted to characterize the impact of boric acid (ba) on the cellular and molecular nature of differentiated rat bone marrow mesenchymal stem cells (bmscs). materials and methods: in this experimental study, bmscs were extracted and expanded to the 3rd passage, then cultured in dulbecco’s modified eagle’s medium (dmem) complemented with osteogenic media as well as 6 ng/ml and 6 μg/ml of ba. after 5, 10, 15 and 21 days the viability and the level of mineralization was determined using mtt assay and alizarin red respectively. in addition, the morphology, nuclear diameter and cytoplasmic area of the cells were studied with the help of fluorescent dye. the concentration of calcium, activity of alanine transaminase (alt), aspartate transaminase (ast), lactate dehydrogenase (ldh) and alkaline phosphatase (alp) as well as sodium and potassium levels were also evaluated using commercial kits and a flame photometer respectively. results: although 6 μg/ml of ba was found to be toxic, a concentration of 6 ng/ml increased the osteogenic ability of the cell significantly throughout the treatment. in addition it was observed that b treatment caused the early induction of matrix mineralization compared to controls. conclusion: although more investigation is required, we suggest the prescription of a very low concentration of b in the form of ba or foods containing ba, in groups at high risk of osteoporosis or in the case of bone fracture.

Exogenous Nitric Oxide Induced Early Mineralization in Rat Bone Marrow Mesenchymal Stem Cells via Activation of Alkaline Phosphatase

Background: Since the low concentration and short-time treatment with sodium nitroprusside (SNP), a nitric oxide (NO)–donor, cause no harm to rat bone marrow mesenchymal stem cells (MSCs), we studied the impact of SNP on MSCs differentiation. Methods: MSCs were treated with 100 and 1000 µM of SNP for 1 hour in every 48 hours and after 5, 10, 15, and 21 days in osteogenic media. The viability an...

بررسی آزمایشگاهی اثر ویتامین E بر قدرت زیستی، تغییرات مورفولوژیک و القاء تمایز استئوژنیک در سلول‌های بنیادی مزانشیمی مغزاستخوان رت بالغ

Introduction: Vitamin E as a strong antioxidant plays an important role in inhibiting free radicals. Therefore, this study aimed to investigate the effect of vitamin E on the viability, morphology and osteogenic differentiation in bone marrow mesenchymal stem cells of an adult rat. Methods: The bone marrow mesenchymal stem cells were extracted using the flashing-out method. At the end of the t...

Sodium Arsenite Caused Mineralization Impairment in Rat Bone Marrow Mesenchymal Stem Cells Differentiating to Osteoblasts

ABSTRACT Background: Sodium arsenite (SA) recently has been recommended to be used in malignancy therapy. Our studies showed, SA in short and long period of treatment caused reduction of rats Bone Marrow Mesenchymal Stem Cells (MSCs) viability and induced caspase dependent apoptosis. The aim of this study was to investigate the effect of SA on osteogenic differentiation of MSCs. Methods: MSCs...

Myo-inositol at High Concentration Reduced Viability and Proliferation of Rat Bone Marrow Mesenchymal Stem Cells via Electrolyte Imbalance and Elevation of Aerobic Metabolism

Myo-inositol (MI) which is produced at low concentration is an essential substance for animal’s natural growth. This study was performed to investigate the effects of MI on viability, proliferation and some biochemical factors of rat bone marrow mesenchymal stem cells (BMSCs). To investigate the cell viability using trypan blue assay, BMSCs after third passage were treated with different concen...

The Consequence of Vitamin E Exposure on In Vitro Cadmium Toxicity in Rat Bone Marrow Mesenchymal Stem Cells

This investigation aimed to examine the protective function of vitamin E on rat bone marrow mesenchymal stem cells (MSCs) treated with cadmium chloride. Rat bone marrow MSCs were extracted using flashing-out and cultured in DMEM containing 10% FBS and 100 U/ml Pen/Strep. At the end of the third passage, cells were divided into 4 groups including control, cadmium chloride, cadmium chloride + vit...
